Formation of the ¥á1-Adrenoceptor-G-protein complex by stimulation with ¥á1-agonist in pig liver

Abstract
One of primary functions of the is to modulate calcium levels in cells. In transmembrane signaling transduction, the ¥á1-Adrenoceptor activates a 69 kDa phospholipase C through novel Gh-proteins, which are suggested species specific. In the present study, a ternary complex of ¥á1-agonis-adrenoceptor-G-protein was developed by stimulating membrane fractions of pig liver with ¥á1-agonist, epinephrine, and then purified with heparin-agarose and wheat germ agglutinin-agarose affinity chromatography. Inclusion of ¥á1-Adrenoceptors and 77 kDa Gh-like proteins in the ternary complex were confirmed by [3H]prazosin binding assay and photoaffinity labeling with [ -33P]GTP. The ternary complex was formed by stimulating with ¥á1-agonist in pig liver. These results suggested that Gh-like proteins played roles as signal mediators of ¥á1-adrenoceptor.
